ABSORPTION OF VITAMIN B_(12) FROM RAT¢¥S INTESTINAL RINGS AND OXYGEN CONSUMPTION



Abstract
The present study was aimed to observe the mode of absorption of vitamin B_(12) from the rat¢¥s ileum in vitro under various experimentall conditions. The rat¢¥s ileum (lover third of intestine) was removed by stripping from the mesentery and everted with a probe. The removed ileal segment was cut into rings of approximately 1mm. in width and the rings were placed in reaction flack of Warburg manometric apparatus which contained glucose added (100mg.%) Krebs-Ringer phosphate buffer solution (KRP solution), and Co^(60)-vitamin B_(12) together with intrinsic factor (homogenate of rats¢¥ gastric mucosa) and /or 0.16 N or sodium bicarbonate or hydrochloric acid, Deer Horn extract. And the effects of Deer Horn extract, alkali and acid on the absorption of vitamin B_(12) and oxygen consumption of rat¢¥s intestinal rings were studied.
The srsults obtained were summarized as follows:
1. The intestinal rings absorbed 6.71% of added vitamin B_(12)(4.8§¶mg) and consumed 54.08. of oxygen in the reaction medium which contained only vitamin B_(12) in KRP solution.
2. The absorption of intestinal rings showed the tendency of decrease in the reaction rings showed the tendency of decrease in the reaction medium which contained intrinsic factor and vitamin B_(12) in KRP solution.
3. In the alkaline reaction medium(pH:8.0) which contained sodium bicarbonate, intrinsic factor, and vitamin B_(12) in KRP sulution the absorption of vitamin B_(12) was markedly depressed with an increased tendency of oxygen consumption.
4. In the acid reaction medium (pH:2.5) which contained hydrochloric acid, intrinsic factor and vitamin B_(12) and oxygen consumption decreased.
5. The absorption of vitamin B_(12) in Deer Horn extract added KRP solution increased markedly with no significant change in the oxygen consumption.
6. Above results indicates that there is no close relationship between the vitamin B_(12) absorption and oxygen consumption in rat¢¥s intestinal rings. Further study will be required to determine the possible effect of Deer Horn in the absorption of vitamin B_(12) in vivo and the mechanism by chich the Deer Horn extract increased the absorption of vitamin B_(12) from intestinal rings in vitro.
